Effect of depletion of Nup133 and Nup43 by antibody addition on spindle assembly on sperm chromatin in Xenopus oocyte extract. Spindles (red) were localized by addition of tubulin-rhodamine and chromosomes stained with DAPI (blue). See Orjalo et al 2006. Mol Biol Cell 17:3806-3808.
Xenopus egg extracts were mixed with sperm chromatin, treated to induce spindle formation, and antibodies added to deplete nucleoporin components. Rhodamine-tubulin was added, the preparations fixed with paraformaldehyde, and spindles centrifuged onto cover slips through a glycerol cushion. Samples were examined using a Zeiss Axioskop microscope. See Fig 6B in Orjalo et al. 2006 Mol Biol Cell 17:3806-3818.
